This tool estimates a protein's molar extinction coefficient at 280 nm from its tryptophan, tyrosine and cysteine content, and the absorbance a 1 g/L solution would give.
This tool estimates a protein's molar extinction coefficient at 280 nm from its tryptophan, tyrosine and cysteine content, and the absorbance a 1 g/L solution would give. Those numbers let you convert an A280 reading into a concentration without running a colorimetric assay, provided you know the sequence.

1 Trp, 2 Tyr, 1 Cys. 8192.5 Da. Extinction coefficient 8480 in both states. A280 of 1 g/L is 1.035
Good to know. Every W, Y and C letter in the sequence counts. A FASTA header line is skipped, and when several records are pasted only the first is read.

Aromatic / cysteine composition
Computed from composition using the Pace et al. (1995) parameters (ε = 5500·W + 1490·Y + 125·cystines), the same method as ExPASy ProtParam. The reduced value applies when all cysteines are free; the cystine value assumes every pair forms a disulfide bond.
